Cloning and nucleotide sequence analysis of Zein promoter

Abstract

Promoter plays an important role in regulating the initiation of time- and tissue-specific gene expression. Zein promoter regulates endosperm-specific expression of the Zein gene in maize.The isolation of Zein promoter would lay the basis for genetic experiments,such as the specific expression of exogenous gene in crop endosperm. About 400 bp DNA fragment of 15 ku β-Zein promoter was amplified by PCR using specific primers from genomic DNA of maize inbred lines Lu 2548. Then the purified product was cloned into pMD18-T vector and sequenced. Sequence analysis indicated that the cloned sequence shares around 96% homologues with 15 ku-Zein promoter(GenBank Accession: M72708). Promoter function prediction and cis-element analysis indicated that the sequence may function as an endosperm-specific promoter. The functional verification test is underway. This research might provide the foundation for endosperm-specific genetic modification in maize and other cereal crops.
